Vancomycin has been studied in combination with Gentamicin and Rifampin for the treatment of Staphylococcus Aureus infections, specifically osteomyelitis. The combination of Vancomycin and Gentamicin has been compared to Vancomycin alone in clinical trials for the treatment of MRSA infections.

Mechanism of action
Vancomycin inhibits bacterial cell wall synthesis by binding to D-Ala-D-Ala peptidoglycan precursors. Gentamicin is an aminoglycoside that binds to the 30S ribosomal subunit, inhibiting protein synthesis. Rifampin inhibits bacterial RNA polymerase, blocking transcription. This triple combination provides synergistic bactericidal activity against difficult-to-treat infections, particularly gram-positive organisms and some gram-negative bacteria.
