Mechanism of action
Sonidegib selectively inhibits Smoothened, a key transmembrane protein in the Hedgehog signaling cascade. By blocking SMO, it prevents downstream activation of Gli transcription factors, thereby suppressing the proliferation of cells dependent on Hedgehog pathway signaling. This mechanism is particularly effective in tumors with constitutive Hedgehog pathway activation, such as basal cell carcinoma.
