Mechanism of action
Pixantrone works by intercalating DNA strands, which prevents the enzyme topoisomerase II from unwinding DNA, ultimately leading to cell death. This mechanism is similar to other anthracyclines, but pixantrone has a more favorable side effect profile due to its reduced cardiotoxicity. Cyclophosphamide is an alkylating agent that adds an alkyl group to DNA, causing cross-linking and DNA damage, which triggers cell death. Vincristine is a vinca alkaloid that binds to tubulin, preventing microtubule formation and disrupting cell division. Rituximab is a monoclonal antibody that targets CD20 on B cells, leading to their depletion. Prednisone is a corticosteroid that suppresses the immune system and reduces inflammation.
