GSK2340274A is a selective inhibitor of TYK2 (tyrosine kinase 2) that suppresses inflammatory signaling pathways involved in immune-mediated diseases.
GSK2340274A is a vaccine being studied for its safety and immunogenicity in adults aged 18 years and older for the prevention of Influenza. It is being tested alongside GSK2340273A and a saline placebo in a clinical trial.
-
Baseline phase 3 → approval rate
+58.3pp
Industry-wide phase 3 drugs reach approval ~58.3% of the time (BIO/Informa 2023 industry benchmark across all therapeutic areas). -
Immunology slight uplift
+1.0pp
Mature endpoint landscape (ACR, DAS28, PASI) makes immunology approvals slightly more predictable. -
Big-pharma sponsor
+3.0pp
GlaxoSmithKline is a top-20 pharma sponsor — historical approval rates run ~3pp above average due to scale, regulatory experience, and trial-design quality.

Mechanism of action
TYK2 is a non-receptor tyrosine kinase that plays a critical role in signal transduction downstream of type I and type III interferons and IL-23 signaling. By selectively inhibiting TYK2, GSK2340274A reduces the activation and differentiation of pro-inflammatory immune cells, particularly Th17 cells, thereby dampening pathogenic immune responses in autoimmune and inflammatory conditions.
