Mineral and strong organic acid addition salts of tetracycline or chlortetracycline are prepared by dissolving the amphoteric antibiotic in methyl "Cellosolve" (Registered Trade Mark), and adding one equivalent of the acid dissolved in water. Specified are salts with hydrochloric, hydrobromic, hydriodic, sulphuric and nitric acids.ALSO:A therapeutically effective composition useful for the preparation of intramuscularly injectable liquid preparations comprises a mixture of an acid addition salt of tetracycline or chlortetracycline (see Group IV (b)) and magnesium chloride. It is preferred to use two molecules of magnesium chloride to one molecule of the antibiotic. Specified acid addition salts are the hydrochloride, hydrobromide, hydroiodide, sulphate and nitrate. The injectable vehicle may be water, propylene glycol or oils such as sesame and peanut oils. Optional ingredients for aqueous injections are (a) a non-toxic, water-soluble organic acid, e.g. citric, gluconic, lactic, ascorbic, maleic, malic, malonic and nicotinic acids, an hydroxy acid, or a compound which on addition of water forms such an acid, e.g. glucurono-lactone; and (b) a water-soluble local anaesthetic, e.g. procaine hydrochloride and other dialkylamino alkanol esters of p-aminobenzoic acid.
