USPTO Abstract

An iron-polysaccharide complex is made by heating a polysaccharide with hydrogen peroxide, and at a pH above 8, with a non-toxic ferric compound and bringing the resulting complex into colloidal solution. The polysaccharide may have been heated under alkaline, neutral or weakly acid conditions with the hydrogen peroxide, prior to effecting complex formation. The product may be dried, and formed into solution with a chelating hydroxyacid. Specified polysaccharides are starch, dextran, levan, insulin, mannan and pullulan; specified ferric compounds are the nitrate, chloride, acetate or ferric ammonium chloride or a ferric oxy-salt. The chelating acid may be citric, tartaric, lactic, glycolic or salicylic acid.ALSO:An iron-polysaccharide complex which has been prepared by reacting, at a pH above 8, a polysaccharide which has been treated with hydrogen peroxide and a non-toxic ferric compound and which has been brought into colloidal suspension, may be used in the preparation of an injectable preparation. In examples, the complex is brought into solution using secondary sodium citrate as a chelating agent, the pH is adjusted to 6.3, the solution rendered isotonic e.g. with NaCl, and then sterilized at 105 DEG C. for 1/2 -hour, yielding a solution containing 100 mg Fe/ml. Specified polysaccharides are starch, dextran, mannan, inulin, pullulan and levan. Other chelating agents are citric, lactic, tartaric, glycolic or salicylic acids.
