A method of quantitatively estimating biologically- active cyclosporines, analogues, derivatives and metabolites by a competitive protein binding assay uses water-soluble binding proteins isolated from the cycloplasm of human and animal cells. Tracers include radioactive atoms, fluorescent molecules, enzymes, and chemiluminescent molecules. Solution and solid phase assay methods are disclosed. Also claimed are the water-soluble binding proteins.
