本发明提供了人甲状旁腺素1-34肽相关肽-Pro-Pro-[Arg 11 ]hPTH(1-34)-Pro-Pro,及其构建方法和生产工艺。本发明所述的人甲状旁腺素1-34肽相关肽-Pro-Pro-[Arg 11 ]hPTH(1-34)-Pro-Pro是一种前体肽,在机体内N端的两个脯氨酸残基会被切除,生成有生物活性的[Arg 11 ]hPTH(1-34)-Pro-Pro。本发明还提供了[Arg 11 ]hPTH(1-34)-Pro-Pro的产生方法。甲状旁腺素1-34肽相关肽的制备包含以下步骤:(a)在适合表达人甲状旁腺素1-34肽相关肽-Pro-Pro-[Arg 11 ]hPTH(1-34)-Pro-Pro的条件下,培养含有权利要求3所述重组质粒pED-4P-[Arg 11 ]hPTH(1-34)的宿主细胞;(b)从培养物中分离出具有Pro-Pro-[Arg 11 ]hPTH(1-34)-Pro-Pro肽片段的融合蛋白;(c)分离纯化出Pro-Pro-[Arg 11 ]hPTH(1-34)-Pro-Pro肽;(d)借助体内的二肽酶IV等酶将Pro-Pro-[Arg 11 ]hPTH(1-34)-Pro-Pro肽N端的两个脯氨酸残基切除,形成[Arg 11 ]hPTH(1-34)-Pro-Pro肽。本发明还提供了人甲状旁腺素1-34肽相关肽基因和氨基酸序列、载体和宿主细胞。本发明可高效简便地生产高纯度、高活力的人甲状旁腺素1-34肽相关肽。
