一种Aβ斑块显像剂前体AV45的含量分析方法,属于仪器分析技术领域。 18 F-AV45能在体鉴别AD病人脑中Aβ斑块病理状态,已用于人体内淀粉样蛋白沉积过程的可视化及定量研究,即将走向临床。要获得放化纯较高的PET显像剂 18 F-AV45,必须高纯度前体AV45。本法色谱条件:WATERS600型高效液相色谱仪,2487紫外检测器,色谱柱InertsilC18柱,5μm,4.6×250mm;流动相:50mM磷酸氢二钠︰乙腈v/v15︰85,加三乙胺0.01%;流速:0.8mL/min;AV45进样体积10μL;柱温:室温;检测波长325nm。本法用磷酸盐作缓冲剂,并添加三乙胺使拖尾现象得到改善,峰对称性较好。与文献方法相比,理论塔板数和分离度有很大提高,各杂质分离较好。用325nm检测使各杂质能有效检测。
