Provided herein are methods for preparing monodisperse oligo (ethylene glycol) reagent compositions comprising reacting a halo-terminated oligo (ethylene glycol) with a hydroxyl-terminated oligo (ethylene glycol); also provided herein are small molecule drugs that are chemically modified by covalent attachment of a water-soluble oligomer obtained from the monodisperse compositions to produce conjugates which can exhibit a reduced biological membrane crossing rate as compared to the biological membrane crossing rate of the small molecule drug not attached to the water-soluble oligomer.
