Propofol induced sedation
Propofol induced sedation is a GABA_A receptor agonist Small molecule drug developed by Centre Hospitalier Universitaire de Saint Etienne. It is currently in Phase 3 development for Sedation for medical procedures. Also known as: PIS.
Propofol induces sedation by enhancing the inhibitory neurotransmission mediated by GABA_A receptors.
Propofol is used to treat various conditions, including perioperative pain management, delirium, and lumbar radiculopathy, often in conjunction with spinal anesthesia or quadratus lumborum block. It functions as a positive allosteric modulator of the GABA-A receptor, an anion channel, which is a small molecule mechanism of action.

Mechanism of action
Propofol acts as a positive allosteric modulator of the GABA_A receptor, increasing the frequency of chloride channel opening and resulting in hyperpolarization of the neuronal membrane. This leads to a decrease in neuronal excitability and an overall sedative effect. Propofol's mechanism of action is thought to be responsible for its anxiolytic, anticonvulsant, and muscle relaxant properties.
