Mechanism of action

Marstacimab-hncq is a human monoclonal IgG1 antibody that binds to and neutralizes TFPI, the primary inhibitor of the extrinsic coagulation cascade. By blocking TFPI activity, the drug enhances thrombin generation within the extrinsic pathway of coagulation. TFPI normally inactivates the protease functions of the FXa/FVIIa/TF complex by binding to and inhibiting the factor Xa active site via its second Kunitz inhibitor domain (K2), which is the specific target of marstacimab-hncq.
