Mechanism of action

Fluorouracil (5-FU) is a prodrug that is converted intracellularly to active metabolites, primarily FdUMP and FdUTP. FdUMP inhibits thymidylate synthase, blocking dTMP synthesis and DNA replication. The drug also incorporates into RNA, disrupting protein synthesis. These mechanisms make it effective against rapidly dividing cancer cells.
