amlodipine, hydralazine, terazosin or hydrochlorothiazide
amlodipine, hydralazine, terazosin or hydrochlorothiazide is a Antihypertensive combination (calcium channel blocker, direct vasodilator, alpha-1 antagonist, thiazide diuretic) Small molecule drug developed by Tulane University School of Medicine. It is currently FDA-approved for Hypertension, Heart failure (hydralazine component). Also known as: Norvasc.
This is a combination of four distinct antihypertensive agents that lower blood pressure through different mechanisms: calcium channel blockade, vasodilation, alpha-1 adrenergic blockade, and diuresis.

Mechanism of action
Amlodipine blocks L-type calcium channels to reduce vascular smooth muscle contraction; hydralazine is a direct vasodilator acting on arteriolar smooth muscle; terazosin blocks alpha-1 adrenergic receptors to promote vasodilation; and hydrochlorothiazide is a thiazide diuretic that reduces blood volume and sodium reabsorption. These agents are typically used individually or in combination to achieve additive blood pressure reduction.
